T细胞TCR多样性与对病毒交叉反应性的关系研究3. 骨髓移植后CMV/EBV复发的T细胞治疗与预后诊断研究; 研究工作:1.解析 T 细胞免疫在急性病毒(流感、新冠)和潜伏病毒(CMV、EBV)感染中的作用机制。(1)研究感染中驱动病毒特异性 T 细胞激活、克隆扩增、TCR 多样性和 T细胞耗竭的分子调控网络,揭示流感(如 H7N9)或新冠感染 T 细胞激活和衰竭的免疫机理。(2)研究 CMV/EBV 感染中病毒特异性 T 细胞与血浆 DNA 的动力学关系,以及 CMV T 细胞记忆通胀(memory inflation)的机制。 2. 高效筛选 T 细胞表位和高功能性 TCR-T,推动复发难治性 CMV 和 EBV相关肿瘤 T 细胞治疗与预后诊断,构建通用型广谱 T 细胞疫苗研发。(1)针对特定 HLA,描绘 T 细胞抗原图谱和主导表位 T 细胞的 TCR,制备筛选用的四聚体和 TCR-T,探索特异性 CMV/EBV T 细胞治疗。(2)结合数据库预测获得的新冠、流感 T 细胞表位,开展 T 细胞表位串联或者多条多肽构建T 细胞疫苗,应用乳酸杆菌载体构建口服通用型流感 T 细胞疫苗。 |
